500 _aThe study was conducted to determine the efficacy of Virgin coconut oil prepared through traditional boiling method in in vitro experiment. Using the agar dilution method. the experiment has six (6) treatments and was laid out in replication design Treatment one (T1) is the positive control (streptomycin), treatment two (T2) is the negative control which was pure MHA, treatment three (T3) for 100 µl VCO, treatment four (14) was 200 µl VCO, treatment five (T5) was 300µl, and treatment six (T6) was 400 µl of VCO A total of 18 MHA tubes were used and put into a test tube rack. Result showed that the volume of 100, 200, 300, and 400 µl of 100% VCO showed no growth of bacteria at 18 hours of Incubation. At 24 hours of incubation, there are bacterial colonies but not easily recognized A1 48 hours of incubation, all of the treatments have well-formed bacterial colonies except for the positive control. This VCO prepared through the traditional boiling method which is predominantly composed of lauric acid has inhibitory action on the growth of Staphylococcus aureus within 18 hours of incubation. It is recommended to the future researcher the use of VCO in in vitro experiment to the bacteria such as Escherichia coli using agar dilution method, use in vivo experiment in an animal infected with Staphylococcus aureus and compare the topical and oral efficacy of VCO against Staphylococcus aureus in vivo experiments.
