| General note | The study evaluated the response of Lakatan and Tumuk to micropropagation using male bud inflorescence as ex-plants supplemented with varying levels of thidiazuron (TDZ) as growth regulator. The study analyzed in-vitro callus formation and shoot regeneration in both cultivars and compared the effect of the different TDZ concentrations on in-vitro callus induction, bud formation, and shoot development. The study was laid out in a two-factorial Completely Randomized Design (CRD) with three replications. The two banana cultivars served as Factor A and four TDZ concentrations as Factor B. This study was conducted from October 2023 to February 2024 at the Banana Tissue Culture and Nursery facility of the Nueva Vizcaya State University, Bayombong, Nueva Vizcaya.<br/><br/>Results show that after four weeks of incubation from initiation stage, both cultivars showed high survival percentages and produced friable and milky white calli indicating high potential for differentiation and regeneration. At the multiplication stage where TDZ was incorporated into the media for the first and second subcultures, the number of buds of Tumuk was significantly increased compared to Lakatan, but the length of shoots of Lakatan was significantly longer than Tumuk. The highest concentration of TDZ produced the highest number of buds in both cultivars. In contrast, the highest number of shoots and the longest length of shoots were obtained at a lower TDZ concentration in both cultivars.<br/><br/> The result of the experiment provides conclusive evidence of the feasibility of the application of the technique particularly the use of the male bud inflorescences for Lakatan and Tumuk as explant in micropropagation. Incorporation of TDZ as a plant growth regulator in the MS media is also shown to be effective for inducing callus and bud proliferation which can later regenerate into plantlets with media modification during subcultures. The initial protocol tested in the study can be further optimized and standardized to enhance in-vitro plantlet production.<br/> |
